چکیده

We have measured the number of fluoride ions bound to one molecule of succinate dehydrogenase (SDH) of submitochondrial particles from porcine kidney cortex. Enzyme activity was studied according to the method of Dixon and the number of fluoride ions bound was established arithmetically. A mixed competitive and noncompetitive inhibition of SDH by fluoride was confirmed. At high levels of succinate (~20 mM) that saturate the active site of the enzyme, the binding of one fluoride ion outside the site results in noncompetitive inhibition. At lower levels (~1 mM), the site is not saturated and inhibition is competitive, with two fluoride ions binding to each enzyme molecule.

Studies on the binding of carboxin analogs to succinate dehydrogenase.

Carboxin (5,6-dihydro-2-methyl-l,4-oxathiin-3-carboxanilide) and a number of other carboxanilides are known to be potent inhibitors of succinate oxidation in membrane preparations from animal tissues, fungi, and bacteria, acting at the succinate dehydrogenase-coenzyme Q junction. The kinetics and the maximal extent of inhibition of succinate oxidation by carboxanilides and by thenoyltrifluoroac...

The Reaction of IV-Ethylmaleimide at the Active Site of Succinate Dehydrogenase*

Since 1938 mammalian succinate dehydrogenase has been thought to contain thiol groups at the active site. This hypothesis was questioned recently, because irreversible inhibition by bromopyruvate and ZV-ethylmaleimide appeared not to satisfy the requisite criteria for reaction at the active site. These recent observations of incomplete inactivation of succinate dehydrogenase by N-ethylmaleimide...

Reactive oxygen species level, mitochondrial transcription factor A gene expression and succinate dehydrogenase activity in metaphase II oocytes derived from in vitro cultured vitrified mouse ovaries

The aim of this study was to evaluate the effects of ovarian tissue vitrification and two-step in vitro culture on the metaphase II (MII) oocyte reactive oxygen species (ROS) level, mitochondrial transcription factor A (TFAM) expression and succinate dehydrogenase (SDH) activity. After collection of neonatal mouse ovaries, 45 ovaries were vitrified and the others (n = 45) were...
